Double-label immunohistochemistry to assess labyrinth structure of the mouse placenta with stereology

Summary: for many years the assessment of the mouse labyrinth zone has been performed using Ultrathin Resin Toluidine Blue sections, a technique that is difficult to perform as it requires specific tools and reagents. Because of it, many labs have not performed analysis of the labyrinth zone in their animal model; limiting our knowledge on the biological phenotypes of the labyrinth zone and mechanisms behind of fetal growth restriction and placental insufficiency. The novelty of this work resides on the demonstration that stereological analysis of double staining for Lectin-Cytokeratin sections is as effective as the Ultrathin Resin Toluidine Blue sections to determine parameters in the labyrinth zone such as maternal blood spaces,fetal capillaries, surface area for exchange or barrier thickness.
